金合欢组织培养技术研究

Studies of Tissue Culture of Acacia farnesiana (L.) Willd

  • 摘要: 以金合欢2 a生优树的带芽茎段为材料,着重研究不同激素浓度及其配比对金合欢组织培养各环节的影响,探索适宜的启动、增殖、生根培养基配方。结果表明:适宜的启动培养基配方为:3/4MS+6-BA1.0 mg·L-1+IBA0.5 mg·L-1;适宜的增殖培养基配方为:MS+6-BA0.5 mg·L-1+NAA0.3 mg·L-1+NaH2PO4 40 mg·L-1,月增殖系数可达3.83;适宜的生根培养基配方为:1/2MS+IBA0.6 mg·L-1+NAA0.2 mg·L-1,生根率可达86%,生根效果较理想。

     

    Abstract: Studies were made of medium formulas bud inducement, sub-culture and rooting culture in order to rapidly propagate Acacia farnesiana. The results showed that in the medium of 3/4MS + 6-BA 1.0 mg·L-1 + IBA 0.5 mg·L-1, the optimal bud inducement was obtained, and shoots were effectively propagated in 3/4MS medium supplemented with BA 1.0 mg·L-1, IBA 0.5 mg·L-1 and NaH2PO4 40.0 mg·L-1. And for rooting, the most effective medium was half-strength MS medium supplemented with IBA 0.6 mg·L-1 and NAA 0.2 mg·L-1, and the rooting rate could reach 86%.
