炭疽菌属植物病原真菌CFEM蛋白的理化性质及特征分析

Analysis of Physicochemical Properties and Characteristics of CFEM Proteins in Plant Pathogenic Fungi of the Colletotrichum Genus

  • 摘要: CFEM蛋白是真菌中特有的一类蛋白,在真菌的生长发育和致病过程中发挥着重要作用。以16个炭疽菌属植物病原真菌的CFEM蛋白为基础开展生物信息学分析,以期明确炭疽菌属植物病原真菌CFEM理化性质的特征。通过Pfam数据库以“CFEM”为关键词搜索获得16个炭疽菌的CFEM蛋白序列;通过SMART检查是否存在CFEM结构域,最终获得CFEM蛋白的序列,然后利用蛋白质数据库、Expasy等在线程序对蛋白序列的理化性质等特征进行分析。结果发现,炭疽菌属CFEM蛋白主要为碱性蛋白、稳定蛋白和疏水性蛋白;亚细胞定位集中在质膜;二级结构组成特征为β折叠较少;保守结构域类型包括CFEM、CFEM+TM、WSC+CFEM三类,均具有CFEM保守结构域,且CFEM保守结构域序列氨基酸长度集中在60-66aa,有7个间隔排列的半胱氨酸残基,具有高度的保守性。为解析炭疽菌属植物病原真菌CFEM蛋白的功能和作用机制提供参考。

     

    Abstract: CFEM proteins, unique to fungi, play a crucial role in fungal growth, development, and pathogenicity. This study conducted a bioinformatics analysis based on CFEM proteins from 16 plant pathogenic fungi belonging to the Colletotrichum genus, aiming to clarify the physicochemical characteristics of these proteins. We searched the Pfam database using the keyword "CFEM" and obtained CFEM protein sequences from 16 Colletotrichum species. The presence of the CFEM domain was verified using SMART, and the sequences of CFEM proteins were finalized. Online programs such as protein databases and Expasy were utilized to analyze the physicochemical properties of these protein sequences. The results indicated that CFEM proteins from the Colletotrichum genus are primarily basic, stable, and hydrophobic proteins, with subcellular localization primarily at the plasma membrane. Their secondary structure is characterized by a relatively low number of beta sheets. The types of conserved domains include three categories: CFEM, CFEM+TM, and WSC+CFEM, all proteins possess conserved CFEM conserved domains, and the CFEM conserved domains sequences typically consist of 60-66 amino acids, including seven evenly spaced cysteine residues, exhibiting a high degree of conservation. This study provides insights into the function and mechanism of CFEM proteins in plant pathogenic fungi of the Colletotrichum genus.
