油茶卵形家族蛋白的鉴定与分析

Identification and analysis of ovate family proteins in Camellia oleifera

  • 摘要: 卵形家族蛋白(OFP)是一类植物特有的转录因子,其部分家族成员对果实形状具有重要影响,对该家族的研究有助于提高油茶(Camellia oleifera)果高与产量。基于油茶基因组、转录组及单细胞转录组数据,利用生物信息学的方法对其OFP进行鉴定与分析。油茶基因组中含有37个CoOFP基因;系统进化树分析表明,CoOFP可以分为4个亚家族、7个分支,其中,II-1、III-1、IV-1分支分别含有3、4、11个CoOFP,分别与BnOFP13_2、SlOFP20、OVATE同源。基因表达分析表明,III-1分支的CoOFP26在花芽、柱头、雄蕊、花药、种子、果皮及根中有表达,在成熟子房的胎座(亚群 4与11)中也有表达;IV-1分支的CoOFP23CoOFP29主要在花芽、柱头、雄蕊、花药、种子及果皮中表达,CoOFP23在成熟子房的心皮内层(亚群1与8)有表达,CoOFP29在亚群 1中有表达。CoOFP26可能参与调控油茶胎座及果皮基—顶轴方向的细胞数量,CoOFP23CoOFP29可能参与调控心皮及果皮基—顶轴方向的细胞数量,CoOFP23CoOFP26CoOFP29可能是调控油茶果高的关键基因。

     

    Abstract: Ovate family proteins (OFPs), part of which are related with fruit shape, are a class of plant-specific transcription factors. Investigation of OFPs is helpful to promote the fruit height and yield of Camellia oleifera. Based on the date of genome, transcriptome and single-cell RNA-seq data, identification and analysis of OFPs were conducted in C. oleifera using bioinformatic approaches. A total of 37 CoOFP genes were identified in the genome data of C. oleifera, and the 37 CoOFP proteins were divided into 4 subfamilies and 7 groups according to the topology of phylogenetic tree. Group II-1, III-1 and IV-1 contained 3, 4 and 11 CoOFP proteins, which were homologues of BnOFP13_2, SlOFP20 and OVATE, respectively. The results of transcriptome expression profiling showed that CoOFP26 of group III-1 expressed in flower bud, pistil, stamen, anther, seed, pericarp and root. CoOFP26 were also detected in placenta (clusters 4 and 11) of mature ovaries. Both of CoOFP23 and CoOFP29 of group IV-1 mainly expressed in flower bud, pistil, stamen, anther, seed and pericarp. CoOFP23 was detected in carpel inner layer (clusters 1 and 8) of mature ovaries. CoOFP29 was also detected in cluster 1. CoOFP26 may be involved in regulation of cell proliferation of placenta and pericarp along proximo-distal axis. Both of CoOFP23 and CoOFP29 may be involved in regulation of cell proliferation of carpel and pericarp along proximo-distal axis. CoOFP23, CoOFP26 and CoOFP29 may be the key genes for regulating the fruit height of C. oleifera.
