Abstract:
Using excellent tender stem segments of
arbequina as experimental materials, the effects of factors such as sterilization of olive explants, basic culture medium, and growth regulators on
Olea europaea tissue culture technology were studied. The experimental results showed that the highest yield 36.15% was achieved when 75% ethanol was used for 30 seconds and 0.1% HgCl
2 10 minutes for explant disinfection treatment. The improved MS medium was suitable for inducing shoot germination from explants. The optimal proliferation medium was the improved MS+6-BA4.0 mg·L
−1+NAA0.5 mg·L
−1+TDZ 0.5 mg·L
−1, with an induction rate of 65.3%. The optimal rooting medium was 1/2MS+ABT 0.6 mg·L
−1+IBA 0.2 mg·L
−1, with a rooting rate of 90%. The optimal concentration for adding activated carbon was 0.5 g·L
−1, and the optimal concentration for adding sucrose was 30 g·L
−1
