非洲堇叶片试管快繁体系技术研究

Study on Leaf Test Tube Rapid Propagation Technology System of Saintpaulia ionantha Wendl

  • 摘要: 以引进的13个品种非洲堇的叶片为外植体,探索了非洲堇外植体的消毒方案和诱导、增殖、生根培养方案,初步建立了非洲堇叶片试管快繁体系。研究表明:0.1% HgCl2作用5 min和10% 84作用8 min,非洲堇无菌外植体获得率分别为67.9%和72.8%;愈伤出芽诱导培养基:MS+蔗糖30 g·L−1+卡拉胶6.5 g·L−1+6-BA 0.15 mg·L−1+NAA 0.1 mg·L−1;较佳增殖配方:MS+蔗糖30 g·L−1+卡拉胶6.5 g·L−1+6-BA 0.2 mg·L−1+NAA 0.1 mg·L−1+IBA 0.15 mg·L−1,平均有效增殖系数可达6.43;较佳生根配方:1/2MS+蔗糖15 g·L−1+卡拉胶6.0 g·L−1+IBA 1.0 mg·L−1+NAA 0.05 mg·L−1,平均生根率92.67%。

     

    Abstract: Taking the leaves of 13 introduced varieties of Saintpaulia species as the explants, the sterilization plan and the induction, the proliferation, the rooting culture plan of Saintpaulia explants were explored, and the leaf test tube rapid propagation system of Saintpaulia was preliminary established. The results showed that the acquisition rate of sterile explants treated by 0.1% HgCl2 for 5 minutes and 10% 84 for 8 minutes was 67.9% and 72.8%, respectively. Medium for callus induction: MS + sucrose 30 g·L−1+ carrageenan 6.5 g·L−1+6-BA 0.15 mg·L−1+NAA 0.1 mg·L-1. Better formula for proliferation: MS + sucrose 30 g·L−1 + carrageenan 6.5 g·L−1 + 6-BA 0.2 mg·L−1 + NAA 0.1 mg·L−1 + IBA 0.15 mg·L−1, and the average effective proliferation coefficient could reach 6.43. The optimal rooting formula: 1/2MS + sucrose 15 g·L−1 + carrageenan 6.0 g·L−1 + IBA 1.0 mg·L−1 + NAA 0.05 mg·L−1, and the average rooting rate was 92.67%.
