Effects of Medium Types and Growth Regulators on Cuttage Rooting of Ormosia hosiei

Abstract
To establish a suitable cutting technology system for Ormosia hosiei in Sichuan, this study conducted a cutting experiment with Sichuan-sourced O. hosiei in Yuchen Mountain, Luzhou City, Lu County, Sichuan Province. The changes of soluble sugar contents and oxidase (PPO, POD, NAAO) activities during rooting were analyzed. The variance experimental design was used to analyze the effects of meduim, growth regulator, growth regulator concentration, and treatment time on cuttage rooting. The results showed that during the rooting process of cuttings in both the growth regulator treatment group and the control group, the levels of soluble sugar contents, PPO and POD activities initially increased and subsequently decreased, while NAAO activities showed an opposite trend. The inflection points for these changes occurred earlier in the growth regulator treatment group compared to the control group. The medium, growth regulators, growth regulator concentration, as well as the interactions between growth regulator and concentration, and between growth regulator and treatment time, had highly significant effects on the rooting rate. The highest rooting rate and average length of new shoots were observed in the pure vermiculite medium, while the largest average number of thick roots was found in the medium with a volume ratio of 75% vermiculite and 25% perlite. Both ABT and NAA resulted in relatively high average rooting rates, with ABT showed greater average lengths of new shoots and fresh weight compared to NAA. Among the ABT treatment combinations, higher concentrations (800 mg/L) with shorter treatment times (10 min, 30 min) yielded higher average rooting rates. Research indicated that the dynamic changes in soluble sugar contents and oxidase activities within cuttings could be expedited through the application of growth regulators, thereby enhancing root formation. Through mean comparison analysis, the optimal combination of factors for O. hosiei cuttings is determined to be the use of vermiculite as the medium coupled with an 800 mg/L ABT treatment for 10 to 30 minutes.
